【摘要】目的探讨兔肝缺血再灌注损伤的机制及N-乙酰半胱氨酸对兔肝的保护作用。方法选40只实验兔,随机分为肝缺血再灌注组(IR组)和肝缺血再灌注+N-乙酰半胱氨酸组(IR+NAC组),于门静脉阻断前(T0),门静脉开放即刻(T1),开放后2h(T2),开放后5h(T3),开放后7h(T4)时间点记录心率(HR),平均动脉压(MAP),分别测定各组血清丙氨酸氨基转移酶(ALT),天门冬氨酸氨基转移酶(AST),丙二醛(MDA),超氧化物岐化酶(SOD)并检测肝细胞凋亡数。结果与T0比较,两组T1,T2,T3,T4时间点ALT,AST及MDA显著升高,SOD明显降低;与IR组比较,IR+NAC组T1,T2,T3,T4时间点ALT,AST及MDA显著下降,SOD明显升高,且IR+NAC组T3,T4时凋亡细胞数减少。结论 NAC对兔肝缺血再灌注损伤有保护作用,可能是由于NAC清除氧自由基,抑制肝细胞凋亡。

[Abstract] Objective To investigate the protection of N-acetylcysteine (NAC) hepatic ischemia reperfusion (IR) injury of rabbits. Methods Forty rabbits were randomly divided into IR group and NAC group. Heart (HR), mean arterial pressure(MAP) , serum level of alanine aminotranserase(ALT), serum level of aspartate aminotransferase (AST), serum level of malondialdehyde(MDA) and serum level of superoxide dismutase(SOD) were determined separately before hepatic ischemia (T0), at the time of the hepatic vascular was released(T1) and the time at 2 h (T2),5h(T3),7h(T4) after hepatic vascular was released. The apoptosis of
liver cell was measured also. pared to T0 in both groups, the serum levels of ALT, AST and MDA at T1, T2, T3 and T4 increased but the SOD level decreased markedly. Compared with IR group , the serum levels of ALT, AST and MDA at T1, T2, T3 and T4 were markedly decreased with the dramatic increase of SOD level in group IR+NAC. The apoptosis of liver cell at T3 and T4 decreased markedly in group IR+NAC, pared to group IR. Conclusion These results suggest that NAC can protect hepatic is
