摘要:本实验主要研究了嗜酸性氧化亚铁硫杆菌在不同碳源、氮源、pH、接种量以及FeSO4初始浓度情况下的生长状况,并且在最优条件下进行生长曲线的绘制。同时对浸矿中菌株使用9k、Leathen、Waksman三种不同培养基进行培养,测定浸矿中菌株的能量来源。实验结果表明,—,以葡萄糖为碳源,以(NH4)2SO4为碳源,FeSO4初始浓度为35g/L-55g/L时,接种量在5%—10%之间时为该菌株最适生长条件。在该条件下对嗜酸性氧化亚铁硫杆菌进行培养,得出其生长曲线见正文。同时,对浸矿中菌株的培养得出以下结论:该浸矿中分离得出的天然菌株无法利用S作为能源物质,但可通过氧化FeSO4获得生长所需能量。在光学显微镜下观察菌株形态,发现其与嗜酸性氧化亚铁硫杆菌形状相同,革兰氏染色呈阴性,可得出其为嗜酸性氧化亚铁硫杆菌。关键词: 嗜酸性氧化亚铁硫杆菌;生长特性;9k培养基10550
Study on the growth characteristics of the Acidithiobacillus ferrooxidans
Abstract: The experiment mainly discuss the growth conditions of Acidithiobacillus ferrooxidans in different Carbon, Nitrogen, pH, inoculum concentration and FeSO4 initial concentration, then draw the growth curve under the optimum condition. Besides, to measure the energy source of the bacterial strain in mineral leaching, employing three kinds of cultures: 9k, Leathen, Waksman to them. The result turns out that the optimum condition is when pH is under the range of -, glucose is the Carbon source, (NH4)2SO4 is the Nitrogen source, FeSO4 initial concentration is under the range of35g/L-55g/L, inoculum concentration is between 5%-10%.Developing Acidithiobacillus ferrooxidans under this condition will work out the growth curve in the text.
