Introduction:
The use of DTPA derivatives has become popular in many areas of chemistry, including medical diagnostics, chemical synthesis, and environmental applications. In this study, we investigated two new DTPA derivatives using ~1H NMR titration.
Experimental:
The two DTPA derivatives were prepared and their ~1H NMR spectra were obtained. The titration experiments were carried out by adding small amounts of a metal ion solution to the DTPA derivatives. The pH was maintained at during the titration. The chemical shifts of the DTPA derivatives were monitored until the saturation point was reached.
Results:
From the ~1H NMR spectra of the DTPA derivatives, we observed resonances for the methylene protons of the DTPA backbone as well as the aromatic protons of the phenyl groups attached to the DTPA. The titration experiments showed that both DTPA derivatives form complexes with the metal ions, and the chemical shifts of the protons on the DTPA backbone and the phenyl groups shift upon chelation.
Discussion:
The chemical shifts of the DTPA derivatives upon metal ion chelation confirmed the formation of complexes. The shifts were more significant for the first few equivalents of metal ion added, indicating strong binding. The pH was maintained at during the titration because it is close to the physiological pH, and we wanted to simulate the conditions under which these compounds may be used in medical applications.
Conclusion:
The ~1H NMR titration experiments showed that both DTPA derivatives form complexes with metal ions. The findings suggest that the DTPA derivatives may have potential applications as chelating agents in medical diagnosis and other areas of chemistry. Further studies to investigate the stability and selectivity of these complexes are warranted.
