Objective: To study the protective effect of Tongnao pill on experimental chronic cerebral ischemic, disscuss the primary mechanism op provide enough evidence for the exploitation of new drug of Tongnao pill by observing the change of the content of GSH-PX and SOD in serum and PDGF-B in brain of experimental chronic cerebral ischemic damage in rats.
Methods: There were 80 healthy wistar rats(250±20g), male and female each half, feeded adaptability after one week. 10 big mice were selected randomly as the blank group at first, use the method of ligate mon carotid arteries to creat rat chronic cerebral ischemic model for the rest 70 surviving rats were feeded recoverably for 3 weeks after operation,then the 40 rats were divided into 4 group randomly, model group, Tongnao pill high dosage group, Tongnao pill low dosage group, and BuChangNaoXinTong group after eed in making group had 10 rats and the number of male and that of female rats were equal, All the rats were administrated once a day for 8 continuous weeks. Take blood from femoral after priming stomach over, then execute the animals and take out brain, get blood serum to measure GSH-PX and SOD, get the brain including intact hippocampal tissue anize fixedly, measure the expression of PDGF-B and pathology observe.
Results: (1).Compared with blank group, GSH-PX in the blood serum of model group was significantly decreased(p<); SOD in the blood serum of model group was increased significantly;PDGF-B in rats, brain tissue was increased in a significant amount(p<). Indicating we eeded in making model.(p<);(2).Compared with model group,
GSH-PX in the serum of Tongnao pill group(high dosage 、 low dosage) and of
BuChangNaoXinTong group were inceased significantly(p<); the contents of SOD in this three group were significantly decreased(p<); the expressiongs of PDGF-B in rats, brain tissue of the three group were increased significantly(p<).Indicating no statistic
