The best basis medium for adventitious buds induction and growth is MS; BA can effectively stimulate adventitious formation on test-tube plant leaves, and the optimum concentration of BA is ·L-1, but KT has no action on it. More buds can be induced on the medium with NAA.
Stage of inducing roots and transplant.
Better stout plantlets are obtained at MS+·L-1+·L-1. In the basis media,B5 is more beneficial than MS and 1/2MS to roots induction as well as growth. NAA is more effective to root induction than IBA. The most suitable medium for roots induction is MS+. The survival rate of transplanting is the relative high after plantlets were transplanted in the soil of one thirth of humus.
Measures of anti-browning
It is knowed that effect of different anti-browning is various to growth of bud from selection of Antioxidantin of Aconitum Sinomontanum. The most suitable anti
-browning is Vc(1000mg·L-1) and cut of explant is superficial brown,bud and leaf is
deep green and growth of its is good. Effect of PA(100mg·L-1)and AC(800mg·L-1) is better for anti-browning. Effect of different anti-browning is various to growth of callus too. The best anti-browning is Vc(400mg·L-1) and the fresh weight ratio of callus is highest,growth of callus is good, callus is yellow-white. Effect of PA (100mg·L-1) and AC(800mg·L-1) is better for anti-browning ,but the fresh weight ratio of callus is low.
