【Abstract】 AIM: To investigate the expression of CD147 gene in human ovarian carcinoma cell lines during hypoxiareoxygenation. METHODS: Ovarian cancer cell lines 3AO and HO8910PM, highly expressed CD147, were incubated in hypoxia incubator and incubated with 50 mL/L CO2 and 950 mL/L N2. After 30min exposure to hypoxia, all of the cells were reoxygenated 4 h and 8 h respectively. Reoxygenation was achieved by replenishing the fresh medium and returning the cell cultures to a normoxic environment. Expression and location of CD147 were assessed by confocal laser scanning microscope (CLSM). RESULTS: CD147 was located at cytoplasm. Hypoxia upregulated the CD147 expression, but reoxygenation decreased the CD147 expression. With reoxygenation for 4 h to 8 h, the levels of CD147 expression dropped to the lowest. CONCLUSION: Hypoxia upregulates the expression of CD147 in human ovarian carcinoma cell lines and reoxygenation downregulates CD147 expression. The upregulated expression of CD147 during hypoxia is reversible.

【摘要】目的:探讨缺氧再氧合过程中CD147基因在卵巢癌细胞系中的表达. 方法:卵巢癌细胞系3AO,HO8910PM分别在混合气体缺氧仓中缺氧30 min后更换新鲜培养液,给予再氧合, 常氧下继续培养0,4,8 h. 激光共聚焦显微镜定位,定量检测CD147的表达. 结果:CD147定位于细胞质. 急性缺氧30 min后两种卵巢癌细胞系中CD147表达量明显高于对照组,再氧合4,8 h时CD147的表达量明显降低. 两种细胞株均表现为再氧合4 h时CD147的表达与对照相
