摘要:目的探讨骨折愈合过程中骨形成蛋白2(BMP2),转化生长因子β1(TGFβ1)mRNA的表达与凋亡发生的相关性及意义. 方法建立颌骨骨折模型,利用原位杂交方法检测骨折愈合的不同时期BMP2和TGFβ1mRNA的表达,同时采用TUNEL法检测细胞凋亡,并进行相关性分析. 结果①骨折愈合早期(5~11d)的间充质细胞、成骨细胞和软骨细胞内有BMP2mRNA表达,而TGFβ1mRNA主要在骨折愈合后期(11~14d)的成骨细胞和软骨细胞内表达;②细胞凋亡在骨折愈合的5d和11d最多,以间充质细胞和软骨细胞为主;③BMP2mRNA的表达与凋亡具有正相关(r=,P<),TGFβ1mRNA的表达与凋亡无相关性(r=,P>). 结论骨折愈合过程伴随凋亡的发生,以控制细胞的数量和清除无用的细胞,BMP2可能直接参与了凋亡发生过程.

Abstract:AIM To elucidate the correlation of bone mor-ic protein2(BMP2),transforming growth factorβ1(TGFβ1)mRNA expression and apoptosis during fracture healing and its An experimental model of fracture was established to detect the expression of BMP2,TGFβ1mRNA and apoptosis in the different stages of healing by using in situ hybridization and TUNEL ①BMP2mRNA was expressed in mesenchymal cells,osteoblasts and chondrocytes on5~11d whereas TGFβ1mRNA in osteoblasts and chondrocytes on11~14d post-fracture;②the apoptotic cells increased markedly on5d and11d,especially in mesenchymal cells and chondrocytes;and③there was positive correlation between BMP2mRNA expression and apoptosis(r=,P<).TGFβ1mR-NA and apoptosis had no correlation(r=,P>0.
