【摘要】目的观察局灶性脑缺血后侧脑室脑室下区N-甲基-D-天冬氨酸(NMDA)受体亚单位NR2B的表达变化。方法 44只雄性SD大鼠,随机分为正常组、假手术组和脑缺血再灌注组,线栓法制作大脑中动脉栓塞(MCAO)90 min再灌注模型,术后分3、7、14、21、28天5个时间点取脑,连续石蜡切片,免疫组化反应、图像分析系统行NR2B阳性细胞灰度值测量。结果脑缺血后,缺血侧NR2B的表达在再灌注后3天开始增加,7天达到高峰(P<),28天降至正常水平(P>);缺血对侧的变化趋势同缺血侧,但表达变化的幅度明显减小。结论局灶性脑缺血后,SVZ区NR2B表达增加,可能参与该区的神经发生。

Abstract:Objective To investigate the expression of NMDA receptor subunit NR2B in the subventricular zone(SVZ)after focal cerebral 44 male SD rats were randomly assigned to 3 groups: normal group, sham-operation group and ischemia/reperfusion group. Transient brain ischemia (90 min) and reperfusion was created by practicing middle cerebral artery occlusion (MCAO). Brain sections cut by paraffin method were processed to test the immunohistochemical reaction of NR2B by SP method and to evaluate the results with LEICA QWin image analysis Significant increase of NR2B positive cells was found in the SVZ on ipsilateral side on 3 d; the expression of NR2B immunoreactive cells reached a peak on 7 d, and then started to decline.
