【摘要】目的检测C3H/HeJ和C3H/SW小鼠感染肠出血性大肠杆菌(EHEC:O157)后肾细胞凋亡,并探讨thl4基因突变对肾细胞凋亡的影响。方法流式细胞术(FCM)检测C3H/HeJ和C3H/SW小鼠感染EHEC:O157后肾细胞凋亡率,。结果 C3H/HeJ小鼠肾细胞凋亡率明显高于C3H/SW小鼠,差异有统计学意义()。结论小鼠感染EHEC:O157后肾细胞凋亡与thl4基因突变相关。

Abstract:Objective To detect apoptosis of renal cells in mice C3H/HeJ and C3H/SW infected with EHEC: O157 and to search for the effect of mutation of gene thl4 on apoptosis of renal The apoptosis of renal cells in mice infected with EHEC:O157 was detected by Flow Cytometry Method (FCM). The results were statistically analyzed by SPSS The apoptosis rate of renal cells in C3H/HeJ mice was faster than that in C3H/SW mice. And the difference was The apoptosis of renal cells in mice infected with EHEC: O157 was associated with mutation of gene thl4.

目前,肾细胞凋亡与肾相关疾病的发生息息相关,肾细胞凋亡率的检测可作为多种疾病诊断的一项指标,其监测可对抵御机体的进一步感染起到重要作用。课题组使用流式细胞术(Flow cytometry method, FCM)检测thl4基因突变的C3H/HeJ小鼠与正常的C3H/SW小鼠的Th1/Th2类细胞因子含量及淋巴细胞亚群含量,结果表明突变小鼠的以上两种指标的含量均高于正常小鼠。本实验利用流式细胞术检测两种小鼠感染EHEC:O157后肾细胞的凋亡率,进一步探讨thl4基因突变与EHEC:O157敏感性的关系。
1 材料与方法
实验动物、主要试剂及仪器和软件
