【摘要】目的: 观察IGFⅡ,IGFⅠR在结肠腺瘤中的表达,探讨其生物学意义。方法: 免疫组化方法分别检测IGFⅡ,IGFⅠR,PCNA在正常结肠黏膜组(A组n=20)、低级别上皮内瘤变组(B组n=50)、高级别上皮内瘤变组(C组n=50)和结肠癌组(D组n=20)中的表达,TUNEL法检测细胞凋亡。结果: IGFⅡ,IGFⅠR,PCNA在结肠癌组和高级别上皮内瘤变组中呈高表达,两者之间无显著差异(),分别与低级别上皮内瘤变组、正常大肠黏膜组相比表达有显著差异();低级别上皮内瘤变组、高级别上皮内瘤变组、结肠癌组的凋亡指数(apoptotic index,AI)逐渐降低,且两两比较有显著差异()。IGFⅡ,IGFⅠR的表达与增殖指数(proliferative index,PI)成正相关、与AI成负相关;PI,AI在低级别上皮内瘤变组IGFⅡ,IGFⅠR阴性和阳性中无显著差异(),在高级别上皮内瘤变组则有显著差异()。结论: IGF
Ⅱ,IGFⅠR的异常表达与结肠腺瘤癌变密切相关;IGFⅡ,IGFⅠR促进细胞异常增殖,抑制肿瘤细胞凋亡,在结肠腺瘤癌变过程中起了重要的作用。

[Abstract] Objective: To observe the expression of IGFⅡand IGFⅠR in colorectal adenoma and investigate its biologial significance. Methods: 140 patients were divided into four groups:20 cases of normal colon mucosa(A group),50 cases of low grade colon intraepithelial neoplasia(B group),50 cases of high grade colon intraepithelial neoplasia(C group) and 20 cases of colorectal carcinoma(D group). The expressions of IGFⅡ,IGFⅠR,PCNA and apoptosis were detected by immunohistochemical and TUNEL.
