【摘要】目的建立一种快速、准确测定人血清中霉酚酸浓度的超高效液相色谱质谱(UPLCMS)测定方法。方法在血清中加入内标吲哚美辛,采用超高效液相色谱联合质谱的方法对血清中的霉酚酸进行分离测定。~ μg/mL的浓度范围内呈良好线性,r=, μg/mL,%,日内、日间变异系数均小于5%。结论该方法准确、快速、灵敏,可用于霉酚酸的治疗药物监测、人体内药代动力学及生物等效性研究。

Abstract: Objective To establish a rapid and accurate method using UPLCMS for quantification of mycophenolic acid in human Indometacin was added to human serum sample and mycophenolic acid in human serum was determined by UPLCMS . Results The calibration curve was linear (r=)among the concentration range of ~
μg/mL and the limit of the quantitative detection was μg/mL. The recovery of the method was higher than %. The coefficients of variation for interday and intraday were both lower than 5%.Conclusion As an accurate, rapid and sensitive method, this assay can be used in the supervision of mycophenolic acid drug and the study of ics and bioavailability.

马替麦考酚酯(mycophenolate mofetil)是霉酚酸(MPA)的2吗啉代乙酯,在体内脱脂化后形成具有免疫抑制活性的代谢物MPA,MPA选择性作用于T、B淋巴细胞,抑制其增殖。临床上用于预防急性器官排异反应,治疗同种异体肾移植后难治性排异反应。霉酚酸的血药浓度测定方法国内外已有报道[17],但以高效液相色谱分离、荧光检测居多,且多数样品处理过程繁琐,分析时间长。本文在参考文献的基础上采用超高效液相色谱质谱(UPLCMS)联用的方法,对血清中霉酚酸成功进行了分离测定。
