【摘要】目的研究Wnt3基因在急性髓系白血病(AML)患者中的表达及其意义。方法应用实时定量PCR(real-time PCR)方法检测10名正常人、40例AML患者中Wnt3基因的表达。结果 10例正常人中检测到较低水平Wnt3基因的表达;Wnt3基因在AML患者中的表达水平增高,AML缓解后Wnt3基因的表达水平较初诊时明显降低;Wnt3的表达与AML患者年龄、性别以及初诊白细胞数等临床特征无关;Wnt3低表达组完全缓解率高于高表达组。结论 Wnt3在AML发病中可能起重要作用,可作为判断AML预后的一个指标。

Abstract: Objective To explore the expression and significance of Wnt3 gene in acute myelogenous leukemia (AML) patients. Methods Expression of Wnt3 gene was detected by real-time PCR in 10 healthy people and 30 AML patients. Results The expression of Wnt3 gene was lower in the 10 healthy people and was higher in AML patients. There was marked decline in the expression level of Wnt3 gene plete remission pared to the initial treatment. The expression level of Wnt3 gene was not correlated with the age, gender and the clinical characteristics at initial diagnosis such as the white blood cell (WBC) count and chromosomal karyotype in AML patients. plete remission (CR) rate in AML patients with low expression level of Wnt3 gene was higher than that in those with high expression level. Conclusion Wnt3 may play an important role in AML pathogenesis and can serve as an index in monitoring AML prognosis.
