【摘要】目的: 研究X射线对肺癌细胞X线修复交叉互补基因1(Xray repair plementing gene 1,1)表达的影响,探讨DNA碱基切除修复机制在肺癌细胞对放疗耐受中的可能作用。方法: 以噻唑蓝还原法(MTT)检测X射线对肺癌细胞(人肺腺癌细胞株 A549)抑制率的影响,实时荧光定量PCR技术检测X1 mRNA的表达水平。结果: 肺癌细胞抑制率多数情况下随X射线照射时间的延长及照射剂量的增大而增加,呈照射时间依赖性(P<)和剂量依赖性(P<);X1 mRNA的表达水平均先增高后降低,4 Gy组和8 Gy组在72 h表达水平最高(P<),16 Gy组和32 Gy组在48 h表达水平最高(P<)。结论: X1 mRNA表达水平的明显改变,提示DNA碱基切除修复机制可能在肺癌细胞对放疗的耐受中起了重要的作用。

[Abstract] Objective: To study the effect of X
ray on expression levels of Xray repair plementing gene 1(1) in lung cancer cells, and explore the effect of DNA baseexcision repair mechanism in the resistance to radiotherapy of lung cancer : Cell inhibition ratio was measured using MTT expression level of 1 mRNA was measured by RFQPCR assay. Results: In the majority, the rate of proliferation inhibiting of lung cancer cells was increased along with the radiation time of Xray prolonging(P<) and the increase of radiation dose(P<).The expression level of 1 mRNA in the lung cancer cells was increased significantly after treatment with Xray, and then highest expression levels of 1 mRNA of 4 Gy group and 8 Gy group were at 72 h(P<), and the highest expression levels of 1 mRNA of 16 Gy group and 32 Gy group were at 48 h(P<) after treatment with Xray. Conclusion: Xray had a fairly obvious function to decreases proliferation of lung cancer cell, and it could induce the increase of the expression levels of 1 mRNA in lung cancer results suggest that DNA baseexcision repair mechanism may pla
