【摘要】目的: 研究X射线对肺癌细胞X线修复交叉互补基因1(Xray repair plementing gene 1,1)表达的影响,探讨DNA碱基切除修复机制在肺癌细胞对放疗耐受中的可能作用。方法: 以噻唑蓝还原法(MTT)检测X射线对肺癌细胞(人肺腺癌细胞株 A549)抑制率的影响,实时荧光定量PCR技术检测X1 mRNA的表达水平。结果: 肺癌细胞抑制率多数情况下随X射线照射时间的延长及照射剂量的增大而增加,呈照射时间依赖性(P<)和剂量依赖性(P<);X1 mRNA的表达水平均先增高后降低,4 Gy组和8 Gy组在72 h表达水平最高(P<),16 Gy组和32 Gy组在48 h表达水平最高(P<)。结论: X1 mRNA表达水平的明显改变,提示DNA碱基切除修复机制可能在肺癌细胞对放疗的耐受中起了重要的作用。

[Abstract] Objective: To study the effect of X
ray on expression levels of Xray repair plementing gene 1(1) in lung cancer cells, and explore the effect of DNA baseexcision repair mechanism in the resistance to radiotherapy of lung cancer : Cell inhibition ratio was measured using MTT expression level of 1 mRNA was measured by RFQPCR assay. Results: In the majority, the rate of proliferation inhibiting of lung cancer cells was increased along with the radiation time of Xray prolonging(P<) and the increase of radiation dose(P<).The expression level of 1 mRNA in the lung cancer cells was increased significantly after treatment with Xray, and then highest expression levels of 1 mRNA of 4 Gy group and 8 Gy group were at 72 h(P<), and the highest expression levels of 1 mRNA of 16 Gy group and 32 Gy group were at 48 h(P<) after treatment with Xray. Conclusion: Xray had a fairly obvious function to decreases proliferation of lung cancer cell, and it could induce the increase of the expression levels of 1 mRNA in lung cancer results suggest that DNA baseexcision repair mechanism may pla
