摘要:目的观察碱性成纤维细胞生长因子(bFGF)对缺氧复氧(A/R)损伤后肌细胞凋亡(apoptosis)相关基因蛋白Bc1-2表达的影响. 方法将培养的骨骼肌细胞分为正常对照组、A/R组及浓度分别为10,20和40μg L-1 的bFGF预处理加A/R组共5组,然后用免疫组化方法(ABC法)进行染色,统计分析Bc1-2阳性的肌细胞百分率、平均吸光度的变化. 结果经bFGF预处理后,Bc1-2阳性的肌细胞百分率(如bFGF20μg L-1 组与对照组相比,%→%,P<)、平均吸光度(如bFGF20μg L-1 组与对照组相比,→,P<)均显著增加. 结论 bFGF对A/R损伤的骨骼肌细胞的凋亡可能具有抑制作用.

Abstract:AIM To observe the Bcl-2expression on the cul-tured skeletal muscle cells in neonatal rats injuried by anoxia/reoxygenation(A/R)and preconditioned by basic fibroblast growth factor(bFGF).METHODS We divided cultured skeletomyocytes into five groups,including control group,anoxia/reoxygenation(A/R)group,bFGF preconditioned with A/R groups(bFGF density was10,20,40μg L-1 re-spectively).Then we used immunohistochemical method(ABC method)to dye culture Being preconditioned by bFGF,Bcl-2positive skeletom-yocytes’percentage rose(such as bFGF20μg L-1 pared to control group,%→%,P<)and average light density increased(such as bFGF20μg L-1 pared to control group,→,P<).CONCLUSION bFGF probably inhibits the occurrence of apoptosis of cultured skeletomyocytes injuried by A/R.
