摘要:目的观察原癌基因表达在哮喘气道重塑中的作用. 方法卵蛋白致敏豚鼠建立哮喘模型,Dot-blot,North-ern-blot分子杂交及免疫组织化学技术观察豚鼠气道上皮、肺组织中原癌基因c-fos,c-myc,c-jun和c-sis的表达及其与气道重塑的关系. 结果正常豚鼠气道及肺组织中c-fos和c-myc mRNA无或很少表达,哮喘发作后,豚鼠气道上皮及肺组织c-fos和c-myc mRNA表达明显增加;免疫组织化学染色显示Fos,Myc,Jun及Sis在正常豚鼠低水平表达,、、嗜酸性粒细胞及中性粒细胞浸润,气道平滑肌显著增生. 结论原癌基因表达在气道重塑过程中有重要作用.

Abstract:AIM To explore the role of proto-oncogenes in the process of airway remodeling in Guinea pigs were used as asthma models challenged by -blot,Northern-blot molecular
hybridization and immunohistochemistry techniques were used to detect the expression of c-fos,c-myc,c-jun and c- c-fos and c-myc mRNA could not be detected or expressed at very low level in the control were greatly increased after the guinea pigs were challenged by after the challenge,the expressions of c-fos and c-myc mR-NA reached the peak and returned to normal4h after the study showed that Fos,Myc,Jun and Sis expressed at low level in control group and increased after ovoglobulin -tochemically positive cells lay in the plasma of airway epithe-lium,in cell nucleus of bronchial epithelium and in inflamma-tory studies showed smooth muscle thick-ened around bronchia and lymphocyte infiltration under mu-cosa or around bronchia smooth Proto-oncogenes expression plays an important role in the process of asthma airway remodeling.

气道重塑(Airway remodeling)是支气管哮喘的重要病理特征之一,、,,控制细胞的生长和分化,-fos,c-jun及Fos,Jun,Myc,Sis的表达,以探讨它们与气道重塑的关系.
1 材料和方法
材料盐酸胍、SDS购自美国Sigma公司,[α-32 P]dATP为北京福瑞公司产品,缺口平移标记盒为BRI公司
