【摘要】目的观察不同浓度的姜黄素对人骨肉瘤U2OS细胞增殖的抑制作用。方法将不同浓度的姜黄素(0、10、20、40μmol/L)作用于人骨肉瘤U2OS细胞12、24、48h,采用四甲基偶氮唑盐(MTT)比色法检测U2OS细胞的增殖。结果姜黄素可显著抑制U2OS细胞的体外增殖,并且随着浓度增加,抑制作用明显增强(10、20、40μmol/L的r值分别为-、-、-,P均<);作用时间愈长,对U2OS细胞的抑制作用也愈明显(12、24、48h的r值分别为-、-、-,P均<), 呈明显浓度和时间依赖性。结论姜黄素能明显抑制人骨肉瘤U2OS细胞的体外增殖,可能为临床治疗骨肉瘤提供了一种新的方法。

ABSTRACT: Objective To observe the inhibition of different concentrations of curcumin on cell growth, and explore the molecular mechanism of curcumin on human a. Methods Curcumin of different concentrations (0, 10, 20 and 40μmol/L) was administered to U2OS cells of human a for 12h, 24h and 48h, respectively. The proliferation of U2OS cells was measured by MTT assay. Results Curcumin significantly inhibited the proliferation of PC3 cells in vitro in concentration and timedependent manners. The higher the concentration of curcumin, the stronger the inhibition was (10
μmol/L, 20μmol/L, 40μmol/L of r=-, - and -, respectively, P<). With the prolonged administration, curcumins inhibition on the U2OS cells became more obvious (Linear correlation analysis showed 12h, 24h and 48h of r=-, - and -, respectively, P<).
