摘要:目的观察小鼠心肌梗死后,骨髓及外周血单核细胞心肌特异性转录因子Nkx2-5的变化。方法用大剂量异丙肾上腺素()连续注射3天造成小鼠心肌急性缺血性梗死模型,第3天注射后30min,通过眼球摘除法取外周血,分离双后肢股骨、胫骨,冲洗骨髓腔得骨髓液,经密度梯度离心分离出骨髓及外周血单核细胞,正常组同心肌缺血梗死组。通过细胞Western blotting分析,比较正常组和心肌梗死组骨髓及外周血单核细胞中心肌特异性转录因子Nkx2-5的变化。结果正常骨髓单核细胞中存在Nkx2-5的表达,正常外周血单核细胞中Nkx2-5无表达;心肌梗死后骨髓及外周血Nkx2-5表达均明显增加,同正常组比较有统计学意义,P<。结论正常骨髓中无需培养或诱导即存在表达Nkx2-5的单核细胞,在心肌急性梗死后向外周血动员增加。

tive in normal bone marrow,but weak or negative in normal Nkx2-5expression became positive and obviously increased in both bone marrow and PBMC after myocardial infarction(P<).Conclusion The mononuclear cells,which express Nkx2-5,reside in bone marrow and are mobilized into peripheral blood after myocardial infarction,participating in repairing the infarction zone.
