摘要:本文基于纳米颗粒比表面积大和有机无机杂化整体基质特殊孔结构和易于制备的特
点,利用戊二醛作为间隔臂固定胰蛋白酶,制备了掺杂 SBA-15 纳米颗粒颗粒有机无机杂化
整体固定化酶反应器。对该酶反应器中 SBA-15 纳米颗粒掺杂量、毛细管长度等制备条件进
行优化后,用标准蛋白牛血清蛋白(BSA)对此酶反应器性能进行评价,在很短时间内,BSA
的氨基酸覆盖度能够达到 47%,且高于 12 h 的自由溶液酶解(34%)并且此酶反应器表现了
良好的重复性和稳定性,还实现了对实际样品牛奶蛋白高效和快速的酶解。

Abstract: In this paper, a anic-anic monolithic immobilized enzyme reactor was
prepared bining advangtages of high specific area of SBA-15 nanoparticles and special
pore structure and easy preparation of hybrid monolithic matrix. After optimization of preparation
conditions including amount of nanoparticles embedded and length of reactor, this enzymatic
reactor was evaluated by standard protein bovine serum albumin (BSA). The results showed that
the sequence coverage of BSA (47%) achieved in a short digestion time was higher than 12 h
in-solution digestion (34%) and this reactor presented good reproducibility and stabilit. It also
achieved fast, high digestion efficiency for real sample milk protein.
