【摘要】目的检测微小染色体维持蛋白5(MCM5)、p16蛋白在肺腺癌组织中表达的改变及其在癌发生发展中的作用。方法采用免疫组织化学SP法检测60例肺腺癌、30例支肺脓肿等非癌病变旁正常肺组织中MCM5、p16蛋白的表达情况。结果 60例肺腺癌组织中MCM5蛋白的阳性表达率为100%,与正常对照组的表达差异有高度显著性(P<)。%(43/60),显著低于100%阳性表达的正常对照组(P<)。肺腺癌组中MCM5、p16蛋白的阳性表达与组织学分级有关(P<);癌组中MCM5蛋白的阳性表达与淋巴结转移有关(P<),而p16蛋白的阳性表达与淋巴结转移无关(P>)。MCM5与p16蛋白的表达与肺腺癌的年龄、肿瘤大小、组织学类型及TNM分期无关(P>)。MCM5蛋白的阳性表达与p16的表达呈负相关(r=-,)。结论 MCM5蛋白的高表达可能涉及了人肺腺癌的发生、发展过程,MCM5蛋白的高表达与癌细胞的增殖、侵袭和转移有关,MCM5的检测可作为评价肺腺癌癌细胞的增殖状态、预测肺腺癌临床预后、指导临床治疗的重要生物学指标。p16基因改变在肺腺癌的发生中起重要作用,p16蛋白是肺腺癌早期诊断的有用指标。

Abstract: Objective To outline the significance of minichromosome maintenance proteins 5 (MCM5) and p16 protein expression and to evaluate their correlations with carcinogenesis and tumor progression in lung adenocarcinoma. Methods The SP immunohistochemical technique was used to detect the expression of MCM5 and p16 in 60 cases of lung adenocarcinoma, and 30 cases of normal lung tissues of non-cancerous lung disease (bronchial pulmonary abscess, etc). Results The expression of positive MCM5 in 60 cases of lung adenocarcinoma was %, it was significantly higher than that in normal lung tissues (). The expression of positive p16 in lung adenocarcinoma was % (43/60), this was significantly lower than that in normal lung tissues (%) (). The expressions of positive MCM5 and p16 protein were associated with histological grading in lung adenocarcinoma (), MCM5 protein positive expression was associated with axillary lymph node metastasis in lung adenocarcinoma () , and p16 protein was not (P>) . MCM5 and p16 protein positive expressions had no correlation with age, tumor size, histological classification and TNM stage (P>). MCM5 was negatively correlated with the expression of p16 (r=-, ).
