摘要:干旱是植物生长和发育的限制因素之一,因此抗旱基因克隆研究非常必要。为了得到
抗旱基因,利用甘露醇模拟干旱胁迫,以发芽为筛选指标从拟南芥突变体库中筛选获得抗旱
突变体 csm1-1。生理生化分析表明,csm1-1 抗旱性与其体内电导率变化、可溶性糖含量升
高和脯氨酸积累有关。利用 TAIL-PCR 技术和 RT-PCR 分析,发现 CSM1 基因(RCI3)编
码一个过氧化物酶。以上发现为进一步研究该基因调控植物抗旱的机理提供了基础。

Abstract: Drought is one of the limiting factors for plant growth and development, and therefore
cloning of novel drought-tolerance genes is very necessary. For obtaining the gene of
drought-tolerant ,The wild-type and mutant lines of Arabidopsis were treated with mannitol to
simulate drought stress to screen for drought -resistant mutants by germination analysis. One
drought-tolerant csm1-1 mutant was isolated. Physiological and biochemical analysis showed that
enhanced drought tolerance of csm1-1 was consistent with lower conductivity,and increased
accumulations of soluble sugars and proline acid. Using TAIL-PCR and RT-PCR, we found that
CSM1 is the RCI3 gene encoding a peroxidase. These results provide basis for further research on
