摘要:目的探讨 miR-22 对头颈鳞癌细胞侵袭转移的影响,并对其分子机制进行初步探讨。
方法利用脂质体介导的 hsa-miR-22 mimic 及 inhibitor(含阴性对照)分别转染头颈鳞癌
Tu686 细胞,划痕实验及 Transwell 侵袭实验检测其体外迁移及侵袭能力的改变,Western blot
检测上皮-间质转化分子标志物的改变。结果 hsa-miR-22 mimic 能抑制 Tu686 细胞的体外
迁移及侵袭能力,相反 hsa-miR-22 inhibitor 能促进 Tu686 细胞的体外迁移及侵袭能力。且
miR-22 过表达后能在 Tu686 细胞中抑制上皮-间质转化的分子标志物的改变,即 E-cadherin
的表达上调和 Vimentin 的表达下调。结论本研究阐明了 miR-22 对头颈鳞癌 Tu686 细胞的
侵袭和转移能力具有调控作用,且该调控作用的发生与 miR-22 对 EMT 的抑制作用密切相
关。
