摘要:聚合物纳米胶束作为药物传输载体具有独特的结构和性能特点,如药物负载率高、体
内循环时间较长、生物毒性低等,可以有效地改善直接用药的种种不足。本文合成了一种新
型的阳离子聚合物载体聚乙二醇- 聚天冬酰二乙烯三胺- 赖氨酸接枝胆酸
(PEG-PAsp(DETA)-LysCA2)。通过 1H NMR、FT-IR 表征了聚合物的结构,O/W 乳液法自主
装(PEG-PAsp(DETA)-LysCA2)空胶束,动态光散射检测胶束的粒径大致为 33 nm,透射电镜
TEM 观察胶束的粒径与 DLS 检测一致。Zeta 电位为+14 mV,虽然相对较低但是也足够用
来复合 siRNA。所以该胶束可以用作联合传输基因药物和化疗药物的多功能药物载体。

A novel multi-functional carrier based-on biodegradable
polymer in tumor therapy

Abstract: Nanoscale polymeric micelle ,demonstrating high drug loading,long physiology
circulation time, low biological toxicity, has achieved attention of many researchers in recent
years. Herein, we developed a new multifunctional delivery system Poly(ethyleneglycol)-poly{N’
-[N- (2-aminoethyl)-2-aminoethyl]aspartamide}-lysine-g-CA micelles for the co-delivery of
siRNA and anticancer drug. The polymer structure was characterized by 1H NMR、FT-IR
respectively. The polymeric micelles were prepared by self-assembly under
ultrasound,
following size analysis by DLS and TEM, which showed the size of micelles is about 33nm. A
available zeta potential can be used to load siRNA via electrostatic interaction.
