摘要
目的
检测 miR-146b-5p 在胰腺癌细胞中的表达,探讨其与胰腺癌细胞侵袭转移
的关系,为胰腺癌发生发展的研究提供新的思路。
方法
1 应用 quantitative Real-time PCR(qRT-PCR)的方法检测 miR-146b-5p 在 6 种
胰腺癌细胞系中的表达情况。
2 在 MIA PaCa-2 细胞系中转染 miR-146b-5p mimics 后检测 miR-146b-5p 的
表达情况。应用划痕实验、Transwell 侵袭实验检测上调 miR-146b-5p 后,MIA
PaCa-2 细胞侵袭转移能力的变化。
结果
和正常胰腺组织相比,miR-146b-5p 在 6 种胰腺癌细胞系中均呈低表达,以
MIA PaCa-2 细胞系最为明显。在 MIA PaCa-2 细胞系中,转染 miR-146b-5p mimics
后,miR-146b-5p 表达增高,上调 miR-146b-5p 后的 MIA PaCa-2 细胞侵袭转移能
力降低。
结论
miR-146b-5p 在胰腺癌细胞中低表达,上调 miR-146b-5p 后 MIA PaCa-2 细
胞侵袭转移的能力受到抑制,有望成为胰腺癌侵袭转移机制研究新靶点。

The suppressive effect of miR-146b-5p on invasion and
migration of human pancreatic carcinoma cells
Abstract
Objective
. The aim of this study is to detect the expression of miR-146b-5p in pancreatic
carcinoma cell lines and investigate the relationship between miR-146b-5p and the
invasion and migration of pancreatic carcinoma cells. In addition it may provide new
ideas for the study of development of pancreatic cancer.
Methods
1 、 The expression of miR-146b-5p in 6 pancreatic carcinoma cell lines was
examined by quantitative Real-time PCR(qRT-PCR) 。
2 、 When MIA PaCa-2 cells have transfected with miR-146b-5p mimics ,the
expression of miR-146b-5p in them was examined by qRT-PCR. Meanwhile, we
investigated the ability of invasion and migration in MIA PaCa-2 cells by scratch
migration assay and transwell invasion assay.
Results
Compared with normal pancreatic tissue, qRT-PCR showed that miR-146b-5p
was poorly-expressed in six pancreatic carcinoma cell lines, especially in MIA PaCa-2.
The expression level of miR-146b-5p was raised after transfected with miR-146b-5p
mimics in MIA PaCa-2 cells, meanwhile the ability of invasion and migration was
down-regulated.
Conclusions
