半枝莲中脂溶性化学成分研究

半枝莲为唇形科黄芩属植物半枝莲(Scutellaria barbata )的干燥全草,
主产于我国大部分地区。味微苦,性凉,归肝、肾、肺经,具有清热解毒、活血
祛瘀、利尿消肿、抗癌等功效。
采用硅胶柱色谱,制备高效液相色谱,Sephadex LH-20 凝胶柱色谱等多种手
段进行分离纯化,从半枝莲乙醇提取物的氯仿部位得到 21 个化合物。通过理化
性质及波谱数据鉴定了 12 个化合物的结构。其中黄酮类化合物 5 个,二氢黄酮
类化合物 1 个,蒽醌类化合物 1 个,酚酸类化合物 3 个,甾体化合物 2 个。结
构为β- 谷甾醇(β-Sitosterol, 1) 、 5- 羟基-7,4′- 二甲氧基黄酮
(5-hydroxy-7,4′-dimethoxyflavone, 2) 、 5- 羟基-7,8,4′- 三甲氧基黄酮
(5-hydroxy-7,8,4′-trimethoxyflavone, 3) 、三裂鼠尾草素(5-Hydroxy-6,7,
4′-trimethoxyflavone, Salvigenin, 4)、汉黄芩素(5,7-dihydroxy-8-methoxyflavone,
wogonin, 5)、2-羟基-3-甲基蒽醌(2-hydroxy-3-methyl anthraquinone, 6)、反式-3-(4′-
羟基苯基)丙烯酸乙酯((E)-3-(4-hydroxyphenyl)acrylic acid ethyl ester, 7)、对羟基
苯甲酸乙酯(Ethyl-4-hydroxy benzoate, 8)、羟基苯甲醛(p-hydroxybenzaldehyde,
9) 、柚皮素(Naringenin, 10 ) 、4′-羟基汉黄芩素(4′-Hydroxywogonin, 11) 、胡萝
卜苷(daucosterol, 12)。其中化合物 2, 3, 6, 7 为首次从黄芩属中分离得到的化合
物,化合物 4 为第一次从半枝莲中分离得到的化合物。

ABSTRACT
Scutellaria barbata , the important Chinese traditional medicine belonging
to the Lamiaceae family, genus Scutellaria, is widedly distributed in most parts of
China. It monly used for antipyretic, antitumor, diureses therapy, liver cancer,
stomach cancer and so on.
The thesis investigated the chemical constituents of Scutellaria
compounds were isolated by methods of silica gel column chromatography and
Sephadex LH-20 column chromatography preparative HPLC and recrystallization.
Structures of the pounds were elucidated on the basis of physicochemical
properties and spectroscopic analyses. As a result, twenty-pounds were
separated and identified twelve of them as: β-Sitosterol, 1);
5-hydroxy-7,4′-dimethoxyflavone,2);5-hydroxy-7,8,4′-trimethoxyflavone,3);5-Hydroxy
-6,7,4′-trimethoxyflavone,Salvigenin,4);5,7-dihydroxy-8-methoxyflavone,wogonin,5);2
