色谱法(chromatography)是用于分离多组分有机混合物的一种高效分离技术,色谱分析技术已成为药物分析学科领域中最基本也是最重要的研究手段和方法,具有广阔的应用前景。海洋真菌及其代谢产物中的某种化学成分是天然药物和天然食品添加剂的重要来源。由于某些有效的成分往往含量较低,并与许多其他化学成分并存,其提取分离是一项繁琐而艰巨的工作。色谱技术的发展与应用,对于各类有机物化学成分的分离、纯化与鉴定工作起着重大的推动作用。本论文实验主要是针对一株海洋真菌的四种培养条件(H1、PDB、GMPY和大米培养基)进行优化,其菌丝体和菌液经乙酸乙酯萃取获得粗样后再进行初步的柱色谱分离与纯化,配以TLC、HPLC/UV检测,以期获得含有目标化合物的培养条件。筛选结果表明:H1培养基和大米培养基的代谢产物丰富,适宜进一步研究。

ABSTRACT
Chromatography is a highly efficient separation technology which is used to isolate anic mixture. Chromatography technology has e one of the most important and fundamental research tools and methods in drug analysis area. It has broad application prospects. The positions of marine fungus and their metabolites is an important source of some natural medicine and natural food additives. Because of the low contents of some active ingredients and co-existence with many other chemical elements, their extraction and isolation is a tedious and difficult task. Development and application of chromatographic technique is playing a significant role in promoting the separation, purification and identification of anic ponents. Experiment of the thesis focuses on the optimization of four culture conditions (H1, PDB, GMPY and RICE) of the marine fungus. The mycelium and broth samples, extracted by ethyl acetate in advance, conduct a preliminary column chromatography isolation and purification, together with the TLC、HPLC/UV detection, in order to obtain the culture conditions of containing the pounds. These results demonstrate that: H1 medium and RICE medium with rich metabolites and containing some of the pounds are suitable for further study.
