文档介绍:1 高效液相色谱法测定青黛中靛玉红的含量【摘要】目的建立测定青黛中靛玉红的含量方法。方法采用高效液相色谱法,以 spherisorb C18 ( mm × 200 mm) 为色谱柱,甲醇拟 mol/L 乙酸铵(65 ∶ 35) 为流动相,检测波长为 280 nm , 流速为 1 ml/min , 柱温 35℃。结果青黛中靛玉红的含量在 ~ μ g/ml 范围内线性良好(r= 5)。平均回收率 % , RSD % 。结论该方法测定简便快速、可靠,可作为青黛中靛玉红含量的质量控制。【关键词】靛玉红; 青黛; 高效液相色谱法 Abstract:ObjectiveTo establish the method of determination of indirubin from Indigo Naturalis. Methods HPLC was employed, which chromatographic column was spherisorb C18 ( mm × 200 mm), mobile phase was methanol 拟 mol/L of ammonium acetate, flow rate was 1 ml/min, detection wavelength was 280 nm and column temperature was 35℃. ResultsA good linear correlationship was obtained when indirubin content ranged from μ g/ml to μ g/ml(r= 2 5). Average recovery was %, RSD was %. ConclusionThis method is simple, rapid, reliable and suitable for assay of indirubin from Indigo Naturalis. Key words: Indirubin; Indigo Naturalis; HPLC 青黛为十字花科植物菘蓝 Isatis indigotica 、爵床科植物马蓝 Baphicacanthus cusia 、豆科植物木蓝 Indigoferas tinctoria 、蓼科植物蓼蓝 Polygonum tinctoriu m 等的叶或茎叶, 经加工制得的干燥蓝色粉末或团块, 是常用中药。近年来研究发现, 靛玉红是青黛中的抗癌有效成分, 主要抑制白血病细胞的 DNA 合成。为了有效控制其质量,制定科学的质量检测标准, 确保疗效, 本文采用高效液相色谱法测定其活性成分靛玉红的含量。 1 仪器与试药 仪器 Shimadzu HPLC 仪, LC 拟 10ATVP 溶液输送泵, SPD 拟 10AVP 紫外可见检测器, SLC 拟 10AVP 控制器, CTO 拟 10ASVP 控温箱, LcSolution 工作站。 试药青黛( 福建仙游), 靛玉红对照品( 中国药品生 3 物制品检定所, 717 拟 200204 ,供定量测定用) ,甲醇为色谱醇;乙酸铵、
