摘要
胰酶是从猪、牛、羊等胰脏中提取的多种酶的混合物,是我国生化制药行业的重要产品之一。本论文研究了絮凝法沉淀分离胰酶。通过实验,筛选出了具有良好絮凝效果的絮凝剂——壳聚糖。探讨了壳聚糖用量、pH值、温度等因素埘絮凝作用的影响,并比较了絮凝胰酶和***胰酶性质的差异。
壳聚糖的用量、pH值和温度对絮凝有明显的影响。增加壳聚糖的用量有利 r絮凝沉淀,但是会造成胰酶比酶活的下降,实验表明壳聚糖的适宜用量为 ;在pH5一-6范围内,絮凝效果好,胰酶比酶活相对较高:升高温度有利于絮凝但会造成酶活损失,故一般选择在20℃左右操作。
用欧洲药店标准Jv方法测定絮凝胰酶的比酶活,结果为蛋白酶1307U/g、、,酶活收率分别为:%、%1N %。其中蛋白酶和淀粉酶的活力较低,与***胰酶(3159U/)相比有较大的差距,但是脂肪酶活力较高,高于***。测定絮凝沉淀胰酶滤液的酶活,结果滤液中酶活所占的比率很低,说明絮凝沉淀比较有效。
经测定,絮凝胰酶的热稳定性显著增强,最适温度比***胰酶提高10。C; 最适pH值向酸性方向移动,对酸性环境的稳定性增)Jib表观米氏常数K。有所增加。说明壳聚糖对胰酶有固定化作用。
