Aim:To investigate the allele frequence of CYP2J2·7 in Chinese healthy Han population,and then determine the influence of CYP2J2·7 polymorphism Oil ics of ebastine and its metabolites in Chinese healthy males.
Methods:Polymerase chain reaction-restriction fragment length polymorphism (PCR-RrLP)was employed in CYP2J2 genotype healthy males were selected to participate in this were CYP2J2·1/·1 wild-type,the others were CYP2J2·7/·l was given a single oral dose of lOmg ebastine and midazolam after a overnight fast,the blood samples were drawn at 0、、、l、、
、2、3、4、6、8、12、24、36、48hours after dmg administration..We use -MS tO detect the concentration of midazolam and ebastine and its rISe midazolam to identify phenotype of CYP3A4,and selected individuals with similar
CYP3A4 phenotype to analyze ebastine metabolism.
Results:101 healthy male volunteers were genotyped for CYP232 polymorphism. The frequencies ofCYP2J2·1 and CYP2J2·7 allele were %and %,respectively. The allele fiequencies were in Hardy-Weinberg ics parameters of carebastine(major metabolites of ebastine)showed some differences between the two genotype AUG一48 and AUCo.,tof carebastine were higher in ·I/·l group than in the CYP2J2·7/·l
group(--4-,-84
,m1).But it showed no difference in statistics。
Condusion:The CYP2J2·7 valiant is mon in Chinese male,which frequency is %.In groups of CYP2J2·7/·l genotype, had the tendency of decreasing,but had not significant difference.
