Protective Effect of PHC on Hypoxia/Hypoglycemia and
Reperfusion Injury of Rat Hippocampal Slices
Abstract
Objective To study the protective effect of PHC on hypoxia/hypoglycemia and reperfusion
injury of hippocampal slices, and reveal the mechanism of this effect,which could
provide scientific evidence for widening its clinical application.
Methods
1 Effects of PHC on hypoxia/hypoglycemia and reperfusion injury of
hippocampal slices
Experimental model of H/R in rats was adopted. Slices equally were divided into
five groups: Control group, H/R group, PHC 2µmol·L-1 (PHC2)group, PHC
10µmol·L-1 (PHC10)group, PHC 50µmol·L-1(PHC50)group. Those indexes
including LDH release rate, tissue injury rate by TTC staining method. The changes
of pathomorphology by HE staining were observed as well.
2 Mechanism of the effects of PHC on hypoxia/hypoglycemia and reperfusion
injury of hippocampal slices
Experimental model of H/R in rats was adopted. Slices equally were divided into
four groups: Control group, H/R group, PHC 50µmol·L-(1 PHC)group,Sco50µmol·L-1
(Sco)group. The expression of pERK1/2 and cPLA2 were detected by Western Blot,
content of amino acids in artificial cerebrospinal fluid of hippocampal slices were
measured by HPLC. The expression of pERK1/2、Bax、Bcl-2 and Caspase-3 in
hippocampal slices CA1 field were detected by immunohistochemical method. Results

LDH release rate of H/R group was ±%,which was more than con-
trol group(±%,P<).Compared to H/R group,PHC 2µmol·L-1,10µmol·L
-1,50µmol/L groups all can reduce LDH release,was %,%(P<),%
(P<)of H/R group effect was strengthened gradually
in dose
