自动生化分析在糖尿病动物模型研究中的应用
摘要
自动生化分析技术可以作为一种有效的工具来比较罗格列酮
和吡格列酮在非胰岛素依赖型糖尿病动物模型上的降糖和降脂作用。方
法:遗传性黄色 KKAy 糖尿病小鼠给予罗格列酮[H4](20mg/Kg 体重/
天)/吡格列酮(100mg/Kg 体重/天)/安慰剂治疗,连续 6 周。采用自动
生化分析仪及配套试剂可直接检测糖尿病小鼠的血糖[H5]、甘油三酯、
高密度脂蛋白胆固醇[H6][H7]、低密度脂蛋白胆固醇等指标。结果:自动
生化分析所得到的高密度脂蛋白胆固醇的浓度与传统方法基本一致,且
性能指标优异。与服用安慰剂组相比,罗格列酮和吡格列酮均能明显降
低血糖,但两者针对血脂的影响存在差异:吡格列酮能明显降低甘油三
酯的含量,而罗格列酮对甘油三酯却有较明显的提高作用,但两者均能
显著增加高密度脂蛋白胆固醇的含量,而对低密度脂蛋白胆固醇的影响
并不明显。动物实验结果与临床实验结果之间具有一定的可比性。结
论:自动生化分析技术可以大大提高临床实验的效率,KKAy 小鼠可以
较好地用来评价罗格列酮和吡格列酮的降糖和降脂效果[H8]。

IN RESEARCH OF DIABETIC ANIMAL MODEL
ABSTRACT
Automatic clinical chemistry techniques could be employed pare
the glycemic and lipid effects of Pioglitazone and Rosiglitazone in animal
models of non-insulin-dependent diabetes mellitus (NIDDM). Methods:
Diabetic animal model of Yellow KKAy mice were treated with
Rosiglitazone( 20mg/Kg body weight/day ),Pioglitazone( 100mg/Kg body
weight/day ) ,or placebo for 6 weeks. The level of serum glucose,
triglyceride,HDL-C,LDL-C before and after treatment was detected directly
with automatic clinical chemistry instruments and relevant reagents. Results:
The concentration of HDL-C obtained by automatic biochemistry analysis is
in accordance with that from conventional method,and the former method
shows better performance. Comparing with placebo,Pioglitazone and
Rosiglitazone both significantly reduced the glucose level,but their effects on
lipid were different: triglyceride level was reduced by Pioglitazone,while was
increased by rosiglitazone. Additionally,both Pioglitazone and Rosiglitazone
could significantly increase HDL-C level,but their effects on LDL-C were
not obvious. The results from animal model parable to tha
