【摘要】目的:研究培哚普利对氧化修饰的低密度脂蛋白(oxLDL)诱导的U937泡沫细胞血管内皮生长因子(VEGF)mRNA、蛋白表达的影响。方法: U937细胞与80 μg/mL oxLDL孵育48 h,建立U937泡沫细胞模型,以不同浓度的培哚普利(、、 μmol/L)预处理U937细胞24 h再加入80 μg/mL 的oxLDL孵育48 h,通过逆转录聚合酶链式扩增反应(RTPCR)和酶联免疫吸附试验(ELISA)分别检测U937细胞VEGF mRNA和蛋白的表达情况。结果: U937泡沫细胞组VEGF mRNA的表达较U937细胞对照组明显增加[(±)vs(±)(P<)],培哚普利干预后,随着浓度(、、 μmol/L)的增加,U937泡沫细胞的VEGF mRNA表达水平呈浓度依赖性下降[(±)vs(±)vs(±)(P<)];U937泡沫细胞组VEGF蛋白表达较U937细胞对照组明显增加[(1 ±) pg/mL vs (±) pg/mL(P< )],培哚普利干预后,随着浓度(、、 μmol/L)的增加,U937泡沫细胞VEGF蛋白表达呈浓度依赖性降低(1 ±) pg/mL vs (1 ±) pg/mL vs (1 ±) pg/mL(P<)]。结论: 培哚普利能够浓度依赖性地下调oxLDL诱导的U937泡沫细胞VEGF mRNA、蛋白的表达。

[ABSTRACT] Objective: To study the expression of vascular endothelial growth factor (VEGF) induced by oxidized low density liprotein (oxLDL) and the inhibitory effects of perindopril on the levels of VEGF protein and mRNA in the U937 foam : The U937 cells were incubated with oxLDL 80 μg/mL for 48 h and a macrophagederived foam cell model was established. The medium was pretreated with perindopril (、、1 μmol/L) for 24 h , incubated with oxLDL 80 μg/mL for 48 transcriptionpolymerase chain reaction(RTPCR) and enzymelinked immuneosorbent assay (ELISA) were applied to determine the expression of VEGFprotein and mRNA in U937 cells and treated U937 : VEGFmRNA levels in U937 cells after incubation with oxLDL 80 μg/mL increased pared with control cells(foam cells vs control cells, ± vs ±, P<).After treated with perindopril(、、 μmol/L), VEGF protein levels were concentrationdepently attenuated(foam cells vs perindopril treated cells, ± vs ±、±、±, P<). VEGF protein levels in the medium after incubation with oxLDL 80 μg/mL increased pared wit
