【摘要】目的研究黄芪注射液调节ERK1/2信号转导通路抗培养乳鼠心肌细胞再灌注损伤的作用。方法采用培养的SD乳鼠心肌细胞缺氧复氧模型,实验分为五组:A组为正常对照组,B、C、D、E组均为缺氧/复氧组,其中各组培养液组成为:B组缺氧液,C组缺氧液加黄芪,D组缺氧液加PD98059,E组缺氧液加黄芪和PD98059。缺氧4h,复氧2 h。比较缺氧/复氧前后培养液中心肌酶含量,并进行心肌细胞外信号调节激酶(ERK)蛋白表达检测。结果缺氧/复氧后对各组细胞培养液中心肌酶含量进行比较,结果显示各缺氧组与正常对照组比较,心肌谷草转氨酶(GOT),乳酸脱氢酶(LDH),肌酸激酶(CK)均有升高(P<)。黄芪缺氧组与缺氧对照组比较,GOT升高减轻(P<)。ERK抑制剂PD98059对黄芪的保护作用有一定削弱,但没有统计学意义。黄芪可使缺氧/复氧引起的心肌细胞ERK表达增加, PD98059对黄芪引起的ERK表达增加没有明显抑制作用。结论黄芪能够减轻缺血再灌注损伤,其机制涉及了ERK1/2信号途径。

Abstract: OBJECTIVE To evaluate the protective effect of astragalus injection through activation of ERK1/2 pathway in cultured cardiac myocytes. METHODS Studies were conducted on cultured cells. Rat ventricular myocytes were subjected to hypoxia 4h and reoxygenation 2h. Astragalus injection was applied to cells during ischaemia/reoxygenation, myocardial enzyme was determined and ERK1/2 expression was detected by Western blot after 120 minutes of reperfusion. RESULTS In cardiac myocytes, incubation with astragalus during ischaemia/reoxygenation attenuated the extent of cell damage. Protective effect of astragalus injection was decreased by PD98059, an inhibitor of ERK1/2 activation,but had no statistical significants. Astragalus activated ERK1/2 in ischaemia/reoxygenation myocytes. CONCLUSION This studiy show
