【摘要】目的探讨肝细胞癌变过程中Rb基因产物(Rb蛋白)的异常表达及其意义。%二乙基亚硝胺(DEN)诱导Wistar大鼠建立肝癌动物模型,应用免疫组化法检测Rb蛋白在大鼠肝脏癌变及转移过程中的表达。结果正常组20只中均见Rb蛋白表达,肝硬化组21只中有18只Rb蛋白过表达,肝癌组(未转移)68只中有37只Rb蛋白过表达,肝癌组(伴转移)18只中有3只Rb蛋白过表达。结论 Rb蛋白的阳性表达率随病变程度的加重而逐渐降低,这辅证了Rb蛋白在肝细胞癌形成过程中发挥抑癌作用,Rb蛋白的失活可能促进肝癌形成,并与肝癌的发展、转移密切相关。

Abstract Objective To discuss the abnormal expression of retinoblastoma protein (Rb) gene product in the hepatocellular carcinogenesis and its % N-nitrosodethylamine (DEN) was applied to induce Wistar rats to form the model of hepatocellular carcinoma (HCC), and the expression of Rb protein in HCC carcinogenesis and metastasis was detected with immunohistochemical 20 rats in control group all showed the expression of Rb protein, 18 rats out of the 21 rats in liver cirrhosis group had the expression of Rb protein as well as 37 rats out of the 68 rats in HCC (without metastasis) group, and only 3 rats out of the 18 rats in HCC (with metastasis) had the expression of Rb Uhe positive expression rate of Rb protein decreases gradually with the aggravation of lesion degree, which may assist that Rb protein play an inhibiting role in hepatocellularcarcinoma, its inactivity may promote the genesis of HCC and is closely related to the p
