【摘要】目的探讨冬眠心肌细胞内p38MAPK、TNF-α mRNA的变化和意义。方法 10例行冠状动脉搭桥术的冠心病患者,术前1周内用多巴酚丁胺超声负荷试验结合多普勒组织成像确定冬眠心肌及正常心肌的位置,搭桥术中根据检测结果进行取材(分别取正常心肌和冬眠心肌),并经电镜证实。Western-blot法检测p38MAPK蛋白表达情况,原位杂交法检测TNF-α mRNA的活性。结果冬眠心肌细胞内p38MAPK、TNF-α mRNA的表达较正常细胞高;冬眠心肌p38MAPK与TNF-α mRNA表达相关(r=, P<)。结论慢性缺血缺氧时,心肌细胞内p38MAPK、TNF-α mRNA表达增加。TNF-α mRNA促进p38MAPK的活化,p38MAPK、TNF-α是冬眠心肌形成的重要分子生物学因子。

Abstract:Objective To study the changes and significance of p38MAPK and TNF-α mRNA in hibernating myocardium (HM).Methods Ten patients with coronary artery disease scheduled for bypass surgery were subjected to preoperative dobutamine stress echocardiography and Doppler tissue image to locate the position of HM. The specimens of normal myocardium (NM) and HM were taken during the operation, and then confirmed by electron microscopy. p38MAPK and TNF-α mRNA were detected by using Western blot and in situ hybridization The expressions of p38MAPK and TN
F-α mRNA in HM were significantly increased, compared with that in NM. p38MAPK and TNF-α mRNA were positively correlated (r=, P<).Conclusion The expressions of p38MAPK and TNF-α mRNA are increased in ischemic myocardial cells. As TNF-α mRNA promotes the activation of p38MAPK, they two may be important factors in the molecular biology of HM formation.
