【摘要】目的探讨肿瘤坏死因子-α(TNFα)、细胞间粘附分子-1(CD54)在肝纤维化家兔肝移植中的表达及意义。方法普通成年家兔30只,随机分为正常组5只,供体组10只,对照组5只,实验组10只。l4)法建立肝纤维化动物模型。然后,正常组给予假手术,对照组和实验组采用原位肝移植手术。术后24h处死所有实验动物,应用免疫组织化学的方法,检测各组肝脏组织中TNFα、CD54的表达。结果实验组TNFα、CD54的表达均比对照组和正常组明显增加();对照组CD54表达比正常组增加();对照组TNFα的表达与正常组无明显差异(P>)。结论 TNFα、CD54在肝纤维化家兔肝移植中发挥重要作用,可能影响移植肝脏的损伤修复。

[Abstract] Objective To study of the expression and its significance of TNFα and CD54 on liver tissue from liver transplantation rabbit with hepatic Thirty adult rabbits were divided randomly into four groups,five rabbits were in ten normal group (NG),ten in the donator,five in
the control group (CG),and ten in the experimental group (EG).Carbon tetrachloride was injected intraperitoneally to make the hepatic fibrosis model of the rabbit in NG was given sham operation,CG and EG were given liver transplantation operation 24 hours,all of the rabbits were executed,and immunohistochemistry staining was adopted to observe the positive expression of TNFα and CD54 molecular in liver The positive expression of TNFα and CD54 molecular in EG were significantly higher than that in NG and CG(P<),and the positive expression of CD54 molecular in CG was higher than that in NG (P<),but the expression of TNFα in CG was similar as in NG (P>).Conclusion The positive expression of TNFα and CD54 on liver tissue from liver transplantation rabbit with hepatic fibrosis has important effect,which possibly impacts the reparation of transplanted liver.

肝移植已成为治疗终末期肝病的唯一有效手段,但相对于其他外科手术来说,其风险仍然很大。排斥反应及原发性移植肝无功能已成为肝移植手术失败的主要原因。目前,国内外的研究方向主要是肝移植后的缺血再灌注损伤及移植排斥反应。本实验通过建立肝纤维化家兔模型,再施行肝移植,探讨肝纤维化家兔血液中的细胞因子对移植肝脏的作用。
1 材料与方法
