【摘要】目的建立了测定鲜人参中多糖含量的分析方法。方法用苯酚硫酸法,在波长490 nm处测定吸光度,人参多糖在10~160 μg范围内具有良好的线性关系, 7。%,RSD=%。结果人参多糖的最佳提取条件为:料液比为1∶10,提取温度为100℃,提取时间为4 h,, g/100 g。结论该方法简便、准确率高、重现性好。

人参为五加科植物Panax ginseng 的干燥根,在我国有着悠久的药用历史,主要含有人参皂苷、人参多糖、人参多肽及挥发油、多种氨基酸、脂肪酸及微量元素等有效成分。人参多糖对机体的免疫功能、造血功能以及在抗肿瘤和降血糖等方面均有较为肯定的药理学作用。由于人参多糖的来源不同,所以同一植物不同的提取方法获得的多糖,甚至人参多糖的不同组分的生物学活性差异甚大[1~5]。
本实验利用多糖被浓硫酸水合产生的高温迅速水解,产生单糖,并迅速脱水生成醛糖衍生物,在这种强酸条件下与苯酚反应生成橙色衍生物,该衍生物在波长490 nm 处和一定浓度范围内,吸光度与糖浓度呈线性关系,采用比色法对鲜人参多糖含量进行测定。
