【摘要】目的内皮前体细胞(endothelial progenitor cells, EPCs)是实体肿瘤血管新生及肿瘤生长的重要参与细胞,而手术有动员EPCs作用。本研究观察手术对EPCs数量及肿瘤生长的影响。方法Wistar大鼠行腋窝皮下接种Walker 256肉瘤细胞。手术前和手术后14天,测量荷瘤鼠肿瘤大小,流式细胞术分析外周血EPCs数量及ELISA检测血清中血管内皮生长因子(vascular endothelial growth factor,VEGF)水平。结果Wistar大鼠在接种Walker 256细胞后,按肿瘤大小分组发现,肿瘤生长较快组CD34+细胞数量增加明显(P<),KDR+、CD34+KDR+细胞数量虽有增加但不明显。手术后,KDR+ 、CD34+KDR+细胞数量增加(P<),但肿瘤大小无明显差异。此外,未观察到VEGF在肿瘤的生长及手术动员EPCs中的变化。结论干细胞与肿瘤生长有一定相关性。手术能动员EPCs,但在术后短期内,所动员的EPCs对肿瘤生长影响不明显,长期影响有待进一步研究。

[Abstract]ObjectiveEndothelial progenitor cells (EPCs) are important cells involving in solid tumor angiogenesis and tumor growth. Moreover,EPCs can be mobilized by surgery. In this study,we observed effect of surgery on tumor growth and EPCs number in tumor bearing rats inoculated under the armpit with walker 256 a cancer cell line. The tumor size,the number of EPCs and vascular endothelial growth factor (VEGF) level in serum of
rats in each group were observed by flow cytometry and ELISA before and after surgery Wistar rats inoculated with walker 256 cells,we found that tumor growing faster group increased significantly in the number of CD34+ cells (P<) and the number of KDR+,CD34+ KDR+ cells also increased but had not significantly which grouped according to tumor size. After surgery,the number of KDR+,CD34+ KDR+ cells increased (P<) and there was no significant change in tumor size. Moreover,the effect of VEGF was not observed in tumor growth and mobilization of EPCs with may be involved in tumor growth and surgery can mobilize EPCs into blood. However, in the short term after surgery,the effect of mobilized EPCs has no significant on tumor growth and should be studied further for long-term effects.
