【摘要】目的: E2Z)生长、增殖能力的影响,并探讨其可能机制。方法:用MTT及平板克隆形成实验、Western E2Z后增殖能力、EGR1、p53、p16、Cyclin D1的表达。结果:E2Z细胞的生长增殖能力,同一时间点各实验组细胞抑制率相比差异有统计学意义,抑制率与H2O2浓度间表现出明显的剂量效应关系。E2Z无EGR1、p16蛋白表达,可见p53、Cyclin D1蛋白表达;诱导培养后EGR1、p53、 Cyclin D1蛋白表达明显增加。结论:E2Z细胞生长,其作用可能与EGR1及p53蛋白表达增强有关;E2Z中具有可诱导性,在肿瘤治疗中有一定的应用前景。

【Abstract】 Objective: To observe the effects of H2O2 on the proliferation of nasopharyngeal carcinoma cell line (CNE2Z) and study the possible mechanisms. Methods: E2Z cells were treated with H2O2; the assays of MTT and plate colony formation were used to
measure the ability of proliferation E2Z cells; the western blot was used to detect the protein levels of Egr1, p53, p16 and cyclin D1. Results: In a dosage dependent manner, H2O2 inhibited the proliferation E2Z cells, shown by both the MTT and plate colony formation assays with stronger displaying in the plate colony formation assay; without treatment of E2Z cells express slight p53 and cyclin D1 proteins but no detectable Egr1 and p16 proteins; after treatment with H2O2, CNE2Z express significant higher proteins of Egr1, p53 and cyclin D1 with no detectable change p16 protein. Conclusion: H2O2 might inhibit the proliferation E2Z through increasing the expressions of Egr1 and p53 proteins; since Egr1 expression was inducible, Egr1 migh
