摘要:目的观察抗氧化肽 SS-31 对糖尿病小鼠肾脏细胞凋亡的影响。方法雄性 CD-1 小鼠
40 只,采用单一剂量 STZ(150 mg/kg)腹腔注射诱导建立小鼠Ⅰ型糖尿病模型,随机分为:
单肾切除对照组(Control)、单肾切除糖尿病模型组(DM)、单肾切除糖尿病 SS-31 干预
组(SS-31)。采用原位末端转移酶标记技术(TUNEL)检测细胞凋亡; 组织化学染色检
测 Bcl-2、Bax 等在肾组织的表达;Western blot 检测 Bax、Bcl-2、ERK1/2、p- ERK1/2 的表
达。结果与单肾切除对照组相比,单肾切除糖尿病模型组肾脏细胞凋亡明显增加,p- ERK1/2
表达增高,Bax/Bcl-2 比率明显升高。SS-31 干预能够显著抑制糖尿病小鼠肾脏细胞凋亡,下
调 p- ERK1/2 的表达,减少 Bax/Bcl-2 比率。结论 SS-31 能够抑制糖尿病小鼠肾脏细胞凋亡,
抑制 ERK1/2 信号通路激活。

Abstract: Aim To investigate the effect of anti-oxidant peptide SS-31 on kidney cells apoptosis
in diabetic mouse. Methods CD-1 male mice were divided randomly into 3 groups:
uninephrectomized control group(control), uninephrectomized diabetes group (DM), and
uninephrectomized diabetic SS-31 group(SS-31). Apoptosis of kidney cells was analyzed by
DeadEnd™ Fluorometric TUNEL System. The expression levels of Bax, Bcl-2 were observed by
