摘要:本研究探讨亮氨酸对卫星细胞 mTOR 信号通路关键激酶的影响。在提取的肌源卫星
细胞中加入亮氨酸,采用免疫印迹的方法,考察了 2m mol/L 亮氨酸对卫星细胞 mTOR、S6K、
4EBP1、TSC2 和 Akt 的蛋白磷酸化水平同时检测了对蛋白质合成和 DNA 合成速率的影响。
结果可知,肌源卫星细胞在加入亮氨酸作用下,mTOR、S6K、4EBP1、TSC2 和 Akt 的磷酸
化明显激活,而采用 mTOR 的特异性抑制剂雷帕霉素处理细胞,能够立刻中止蛋白和 DNA
的合成,此时亮氨酸的激活无效。也就是说,亮氨酸可以通过激活 mTOR 信号通路关键激
酶,从而活化卫星细胞。

Abstract: The effect of leucine on crucial kinases of mTOR signal pathway was investigated. 2m
mol/L Leucine was added in satellite cells from pig, and western blotting was chosen to illuminate
the influences of leucine on phospharylation of mTOR、S6K、4EBP1、TSC2 and Akt, at the same
time, protein synthesis and DNA proliferation were conducted. Results shown, phospharylation of
mTOR、S6K、4EBP1、TSC2 and Akt were significantly increased, Rapamycin as the inhibitor of
mTOR was added in satellite cells, protein synthesis and DNA proliferation were all terminated in
spite of activation of leucine. In a word, leucine can activate satellite cell by activating crucial
kinases of mTOR signal pathway.
