摘要:目的唾液腺腺样囊性癌是唾液腺中第二常见的恶性肿瘤,其细胞内过表达 Bcl-2 和
Mcl-1 能够抑制凋亡,降低治疗效果。本研究旨在探讨 Bcl-2 拮抗剂:Obatoclax 在唾液腺腺
样囊性癌细胞中诱导自噬与凋亡的关系。方法通过 western blot 和透射电镜检测 Obatoclax
刺激后 ACC-M 细胞自噬水平的变化,然后通过药物(3-MA 和 CQ)抑制细胞内自噬后,
使用 MTT 法观察 ACC-M 细胞活力以及 western blot 检测 caspase-3 裂解情况。结果 western
blot 和透射电镜结果显示 Obatoclax 能促进 LC3-I 向 LC3-II 转化,细胞内部出现大量自噬体
结构。药物阻断 Obatoclax 诱导的自噬后,其细胞毒性明显增加,并且促进 caspase-3 裂解,
最终导致细胞凋亡。结论本研究证明 Obatoclax 能够在人唾液腺腺样囊性癌 ACC-M 细胞中
诱导自噬,并且证实其诱导的自噬具有抑制凋亡的作用。

Abstract: Objective salivary adenoid cystic carcinoma (ACC) of the salivary glands is the second
mon malignancy in salivary gland. However, over-expression of Bcl-2 and Mcl-1 in the
ACC cells leads to inhibition of apoptosis, which reduces the anti-tumor effect of the treatments.
This study was designed to study the relationship between autophagy and apoptosis induced by the
