摘要:目的:研究血管紧张素 II 对小鼠成骨细胞线粒体功能的影响。方法:体外培养小鼠
原代成骨细胞,用 10-6M Ang II 刺激成骨细胞 24 小时,利用线粒体荧光探针测量线粒体
活性氧的产生,并检测线粒体氧化呼吸链酶活性,线粒体膜电位以及 ATP 的合成,确定 Ang
II 对小鼠成骨细胞线粒体功能活性的影响。结果:在经过 10-6M Ang II 刺激后,小鼠成骨
细胞线粒体内活性氧生成明显增加,氧化呼吸链酶活性受到抑制,线粒体细胞膜电位降低,
ATP 合成量下降。结论:Ang II 对小鼠成骨细胞线粒体功能活性有着明显的抑制作用,为
进一步研究高血压与骨吸收疾病相关性机制提供了新思路。

Abstract: Objective :To analyze the effects of Angiotensin(Ang) Ⅱon mitochondrial functions
in primary mouse osteoblast culture. Methods: The mouse osteoblasts were cultured in vitro, and
treated with or without 10-6M AngⅡfor 24 hours. The generation of reactive oxygen species
(ROS) was detected by fluorescent probe MitoSox. The enzymatic activities of mitochondrial
respiratory plex, mitochondrial membrane potentials and ATP generations were also
evaluated to determine the effects of Ang II treatments on the mitochondrial fu
