【摘要】目的: 初步探讨人类β1,3半乳糖基转移酶7(β3GalT7)基因对HL60细胞的细胞周期、侵袭能力等方面的影响。方法: 通过脂质体介导将本实验室构建好的β3GalT7真核正义表达载体pEGFPC1T7Sense和反义表达载体pEGFPC1T7AntiSense转染入HL60细胞;流式细胞仪分析各细胞株的细胞周期改变;Transwell侵袭实验检测β3GalT7对HL60细胞侵袭转移能力的影响。结果: 随着β3GalT7 表达的增高,G2+S期细胞增多,且细胞侵袭能力增强;而转染反义表达载体pEGFPC1T7AntiSense的结果是相反的。结论: 过度表达β3GalT7的HL60细胞株体外增殖和侵袭能力增强。

[Abstract]Objective: To explore the effects of β3GalT7 gene on cell cycle, invasive potential of HL
: Transfect the binant plasmids pEGFPC1T7Sense and pEGFPC1T7AntiSense into HL60 with cycle and invasive potential of the transfected cells with flow cytometry and transwell plate,respectively. Results: As the upregulation expression of β3GalT7, the cell growth rate increased. At 48 h after transfection, significantly increased invasiveness was noted in HL60 transfected with pEGFPC1T7Sense. Conclusion: Overexpressing β3GalT7 in HL60 enhanced proliferative and invasive potential.
