【摘要】目的探讨益肾胶囊对狼疮性肾炎(lupus nephritis, LN)大鼠的治疗作用及其作用机制。方法观察益肾胶囊组大鼠肾功能、24h尿蛋白(PRO/24h)的变化,分别采用双抗体夹心酶联免疫吸附法及放射免疫法检测血清Th1/Th2细胞因子(IL2、IL10、TGFβ1、IL6、TNFα)水平的变化和光镜观察肾脏病理形态学的改变。结果经过6周的治疗,益肾胶囊组及强的松组大鼠肾功能、24h尿蛋白及血清IL6、IL10、TGFβ1、TNFα水平与模型组比较均明显降低,IL2明显升高(P&lt;&lt;),且益肾胶囊组大鼠肾功能、24h尿蛋白及血清IL6、TGFβ1、TNFα水平明显优于强的松组(P&lt;&lt;);益肾胶囊组及强的松组肾脏病理改变均有不同程度的改善,两组比较无明显差异。结论益肾胶囊可明显降低LN大鼠24h尿蛋白和改善肾功能,其作用机制可能与调节血清Th1/Th2细胞因子网络平衡及减轻肾脏病理损害有关。

ABSTRACT: Objective To investigate the therapeutic effect of Yishen Capsule on lupus nephritis (LN) in rats and its mechanism. Methods We observed the changes of renal function and proteinuria /24h of LN rats in Yishen Capsule group. At the same time, Th1/Th2 cytokines (IL
2, IL10, TGFβ1, IL6 and TNFα) in blood serum were measured by double antibody sandwichELISA and radioimmunity, respectively. The pathological changes of the kidney were observed by light microscope. Results After six weeks treatment, renal function, proteinuria /24h as well as serum levels of IL6, IL10, TGFβ1 and TNFα were decreased and IL2 was increased significantly in Yishen Capsule group and prednisone pared with those in the model group (P&lt; or P&lt;). Renal function, proteinuria /24h, serum levels of IL6, TGFβ1 and TNFα were better in Yishen Capsule
