摘要
目的通过检测急性心肌梗死大鼠左心室梗死面积及缺血区凋亡细胞来评价法舒
地尔(fasudil)对大鼠急性心肌梗死后心肌的保护作用。
方法①实验以雌性 wistar 大鼠作为研究对象,将其随机分为 3 组,AMI 组:结
扎左冠状动脉前降支(LAD);治疗组:术前 1h 30mg/kg fasudil 腹腔注射,结扎 LAD;
假手术组:仅在 LAD 下穿线但不结扎。②术后 24 小时,运用伊文氏蓝-红四氮唑双
染色法,确定各组大鼠缺血面积和梗死面积。③利用脱氧核糖核昔酸末端转移酶介导
的 dUTP 缺口末端标记(TUNEL)技术检测各组大鼠缺血区凋亡细胞,计算调亡指数。
④免疫组织化学染色法检测缺血区 bcl-2 和 bax 的表达,计算积分光密度值。⑤逆转
录-聚合酶链式反应(RT-PCR)测定各组大鼠缺血区 Rho 激酶 mRNA 的表达,计算
积分光密度值。
结果:①治疗组与 AMI 组比较,梗死面积显著减小(±%和 ±%,
P<),缺血面积无差异。②AMI 组与假手术组比较凋亡指数明显增加(±%
和 ±%,P<),③AMI 组与假手术组比较 bcl-2 表达降低,bax 表达增加,
Rho 激酶 mRNA 表达增加,均为 P<,而治疗组与 AMI 组比较,凋亡指数显著降
低,bcl-2 表达增加,bax 表达降低,Rho 激酶 mRNA 表达降低,均为 P<。
结论:Rho 激酶参与了 AMI 心肌细胞的凋亡,fasudil 的心肌保护作用与其抗心肌
细胞凋亡效应有关。

Fasudil protects the myocardium in rats after acute

Abstract
Objective To study the effects of Rho-kinase inhibitor, fasudil on protection the
myocardium in rats after acute myocardial infarction(AMI).
Methods 29 female wistar rats were subjected to left anterior descending coronary
artery(LAD) ligation and were divided randomly into three groups: sham operation
group(not ligation of LAD),AMI group and treated before surgery the rats were
received normal saline (sham group and AMI group)and 30mg/kg fasudil(treated
group). At 24 hours after coronary ligation, the rats were sacrificed. The infarct size and
ischemia size were determined by evans-bule and TTC double staining. The apoptotic
cardiocyte in the ischemia region was determined by TUNEL assay and the apoptotic
index(AI) were calculated. The bcl-2 and bax protein expressions were measured by
immunohistochemical staining. The Rho-kinase mRNA expression in the ischemia region
