摘要:本文采用常压水提法提取桂花籽粉末多糖,用苯酚-硫酸法制作标准曲线,通过研究料液比、提取时间、提取温度、提取次数等单因素实验对多糖得率的影响来设计正交实验,获得最优化的提取条件,然后再由Fenton体系和邻苯三酚自氧化体系研究桂花籽多糖对羟自由基和超氧自由基的清除率。实验结果表明,当物料比为1:30,温度为90℃,提取时间为4h,提取次数为3次时,从提取率和效益等综合方面来说,是提取桂花籽多糖的最佳条件。桂花籽多糖对羟基自由基和超氧阴离子自由基有明显的清除作用,并且随着多糖质量浓度的增加,清除能力增强,表明桂花籽多糖对羟基自由基和超氧阴离子自由基的清除能力与多糖质量浓度有明显的量效关系,·ml-1时,%,通过相关方程可以得到桂花籽多糖的EC50= mg·ml-1;·ml-1时,%,通过相关方程可以得到桂花籽多糖的EC50=·ml-1。

Abstract:In this paper, in order to get the optimized conditions, the author extracts the seeds of sweet olive powder haride, makes the standard curve by phend-sulfuric acid, designs the orthogonal experiment through studying the yield of haride which is affected by some single factor experiments such as liquid ratio, extraction time, extraction temperature ,the frequency of extraction and so on, and then by the Fenton system and the pyrogallol autoxidation system of seeds of sweet olive seed haride hydroxyl radical and superoxide radical scavenging rate
. According to the experiment, considering the extraction rate and the effectiveness prehensive terms, it is the best extraction condition with the material ratio 1:30, the temperature 90℃,extraction time 4h, and three times for the extraction. The seeds of sweet olive powder haride towards hydroxyl radicals and superoxide anion radical have scavenging effecting obviously, and with the increase of haride concentration, the clearance enhances. It shows that the sweet olive seed harides concentration toward the clearance capacity of hydroxyl radical and superoxide anion radical has significant does-effect relationship when harides concentration , the hydroxyl radical scavenging rate up to %,through the relevant equations can be Sweet Olive seed haride EC50 = mg • ml-1 harides concentration , the hydroxyl radical scavenging rate up to %,through the relevant e
