摘要目的: 建立高效液相色谱法测定胸腺肽α1的氨基酸组成。方法: 采用OPA试剂柱前衍生法测定氨基酸组成。色谱柱:Agilent ZORBAX Eclipse AAA(*150mm,),流动相:A相为40mmol/L磷酸氢二钠的水溶液,, B相为甲醇∶乙腈∶水=45∶45∶10,采用梯度洗脱,流动相B在17min内从0%升至57%;流速:·min-1;检测波长:338nm。结果:~5μmol/ml,r=,本方法重复性和精密度良好(RSD<2%)。结论:高效液相色谱法测定胸腺肽α1的氨基酸组成,方法简便,结果准确。

Determination of Amino position of Thymosin α1 by HPLC

Abstract Objective: To establish HPLC method for determination of Amino position of the Thymosin α1. Method:OPA-reagent pre-column derivatization method was used. column: Agilent ZORBAX Eclipse AAA(*150mm,);mobile phase A: 40mmol/L Na2HPO4,;mobile phase B:CH3OH∶∶H2O=45∶45∶10,the gradient elution method was used that the mobile phase B was increased from 0% to 57% in seventeen minutes; flow rate:·min-1;detector wavelength:338nm. Results: There is a good linear relationship with every amino acid with the range of ~5μmol/ml (r=),the repeatability and precision of the method is good(RSD<2%). Conclusion: The method to determine amino position of Thymosin α1 by HPLC is simple and accurate.

胸腺肽α1(thymosin 1,Tα1)是胸腺产生的一种蛋白质和多肽激素,能刺激T淋巴细胞的成熟。1966年
