摘要:目的研究逆转录酶抑制剂(AZT)对卵巢肿瘤细胞端粒酶活性的影响. 方法采用TRAP-ELISA方法检测卵巢癌HO-8910细胞在AZT作用前后端粒酶活性的变化,以流式细胞仪分析细胞周期的改变. 结果 AZT作用后的HO-8910细胞端粒酶活性明显受抑制,且细胞的G2 /M期DNA含量明显增高. 结论 AZT能明显抑制肿瘤细胞的端粒酶活性.

Abstract:AIM To study the effect of inhibition of telom-erase activity by reverse transcriptase inhibitors(AZT)on o-varian cancer cells in Telomerase activity was determined by TRAP-ELISA in untreated and treated HO-8910cells by AZT,cell cycle phases were analyzed by flow- Telomerase activity of HO-8910cells was significantly inhibited when treated with AZT,and cell cycle of treated HO-8910cells was changed with marked increase in G2 /M AZT can effective-ly inhibit telomerase activity of tumor cells,and represent an effective and novel approach for
treatment of cancer.
0 引言
端粒酶是一种特殊的逆转录酶,以端粒末端富含鸟嘌呤单链为引物,自身RNA为模板,不断合成端粒重复片段序列添加至染色体的末端,维持染色体的稳定性[1] .近年来大量研究表明,人类85%以上的恶性肿瘤都有端粒酶活性表达,而正常体细胞及良性肿瘤中端粒酶活性表达极低或无表达[2] .在卵巢癌细胞中端粒酶活性普遍呈高表达,提示端粒酶激活可能是肿瘤增殖的决定性因素[3] .逆转录酶抑制剂(AZT)是利用核苷类似物和正常单核苷酸竞争与模板RNA结合,阻止反转录过程,[4] .我们通过AZT对卵巢癌细胞株HO-8910细胞的作用,探讨其对端粒酶活性的影响,为卵巢癌及其他肿瘤的靶向治疗尝试新的途径.
