【摘要】目的:研究昆布提取物(TLPE)在体外对耐药细胞BMDC1(ADR+/+)的逆转作用,探讨其逆转肿瘤多药耐药的机制. 方法:应用流式细胞术测定不同剂量TLPE对BMDC1用药前后细胞内蛋白激酶Cα(PKCα)和谷胱甘肽转移酶π(GSTπ)的表达变化,测定细胞周期和凋亡率;应用姬姆萨染色技术观察细胞形态学改变及细胞凋亡情况;应用比色法检测各组细胞ATP酶(ATPase)变化. 结果:TLPE在70~300 mg/L浓度范围内能明显降低BMDC1细胞内PKCα和GSTπ的表达(),并将细胞阻滞于S期,但不诱导细胞凋亡;TLPE对细胞内ATPase无影响(). 结论:TLPE对子宫内膜癌细胞系BMDC1(ADR+/+)具有耐药逆转作用,其逆转作用与其诱导PKCα和GSTπ的表达降低有关.

【Abstract】 AIM: To study the multidrug resistance (MDR)
reversing effect of Chinese drug Thallus Laminariae PE (TLPE) on human endometrial cancer cell line BMDC1 (ADR+/+) in vitro and its underlying mechanism. METHODS: The expressions of PKCα and GSTπ at the level of protein in BMDC1 cell treated with different doses of TLPE were analyzed by flow cytometry. The cell cycle and apoptosis rate of BMDC1 (ADR+/+) cells treated by TLPE were detected. Their morphological changes were observed by Gimsa staining methods. Colorimetry was used to detect the changes of ATPase on BMDC1 (ADR+/+) cells treated by TLPE. RESULTS: After interfered by TLPE (70-300 mg/L), the expressions of PKCα and GSTπ in cells were decreased significantly ().
