【摘要】目的: 应用AdEasy复制缺陷型腺病毒载体系统构建携带hVEGFA基因的重组腺病毒载体。方法: 采用分子克隆技术,hVEGFA质粒中克隆出hVEGFA基因的编码序列,酶切、连接后,插入腺病毒系统中的穿梭载体pAdTrackCMV,获得重组质粒pAdTrackCMVhVEGFA,将其转化含有腺病毒骨架载体pAdeasy1的大肠埃希菌BJ5183,进行同源重组,获得重组子pAdeasy1hVEGFA。鉴定后,将pAdeasy1hVEGFA转染人胚肾细胞(HEK293A细胞),获得含hVEGFA基因的重组腺病毒(rAdhVEGFA)。反复感染HEK293A细胞扩增腺病毒,50%组织培养感染量法(TCID50)检测病毒滴度,荧光显微镜观测绿色荧光蛋白(GFP)的表达。结果: 成功构建出含有hVEGFA基因的腺病毒载体rAdhVEGFA,转染HEK293A细胞后出现绿色荧光蛋白表达,证明腺病毒载体rAdhVEGFA 转染成功,通过反复感染HEK293A细胞获得大量病毒。结论: 成功构建含有hVEGFA基因的腺病毒载体rAdhVEGFA,可为缺血性脑血管疾病的基因治疗提供基因载体。

[Abstract] Objective: To construct the binant adenovirus vector containing hVEGFA gene. Methods: The hVEGFA gene coding sequence was cloned into pAdTrackCMV plasmid to get pAdTrackCMVhVEGFA , which would be transformed petent BJ5183 carried backbone plasmid pAdeasy1 already. The homologous binant adenoviral plasmid was delivered into HEK293A cells after verification. The identified binant adenovirus (rAdhVEGFA) was amplified in HEK293A cells. Viral particle concentration was determined by TCID50. Results: The binant adenovirus vector was constructed essfully. The expression of GFP was observed and the right gene was found. Conclusion: The binant adenoviral vector carrying hVEGFA gene was essfully constructed, and provided the basis of ischemic cerebrovascular disease′s gene therapy.

血管内皮生长因子(vascular endothelial growth factor, VEGF )是Ferrara等在1989年从牛垂体滤泡星状细胞的条件培养基中纯化出来的一种具有肝素结合活性的生长因子,是在胚胎发生和创伤愈合过程中启动血管形成的一个高度特异性的有丝分裂原,能特异性地作用于血管内皮细胞,促进血管内皮细胞增殖[1]。
VEGF治疗缺血性脑损伤的研究具有重要的现实意义。本实验利用重组腺病毒载体AdEasy系统构建含有hVEGFA基因的腺病毒载体rAdhVEGFA,以为缺血性脑血管疾病的基因治疗提供基因载体。
1 材料与方法
材料和试剂
hVEGFA质粒由青岛海洋研究所惠赠。TaKaRa TaqTM、pMDTM 18T载体和T4 DNA连接酶(TaKaRa公司);穿梭质粒pAdtrackCMV、腺病毒骨架质粒pAdEasy1、大肠埃希菌DH5α菌和BJ5183及HEK293A细胞由
